Yes you can, however the fill level is fairly specific. It would be very hard to measure the correct amount without taking the cover on and off several times. We do make custom products like seat topper pads all the time.
What makes our products so comfortable really isn't the beads under your butt, but the chair as a whole, distributing your weight across a very large surface. When seated on a bead filled pad it's not the same as our bean bag products.
IMHO, you would be better off with a dual density foam, ie; 2" of high density foam and 2" of low density for a total of 4". If you get the proper density you will not bottom out nor will it be too hard.
